AB OBJECTIVE: To study the the effect of cryopreservation, storage and
thawing on lymphocytes obtained from asymptomatic HIV+ve patients. It is
planned that the autologous cryopreserved cells will be expanded and
reinfused when the patient progresses towards AIDS, with the aim of
delaying disease progression. METHODS: Blood mononuclear cells were
obtained by leukapheresis and cryopreserved in DMSO. Samples of fresh
and cryopreserved (> 4 months) cells were analysed for lymphocyte subset
proportion by 3 colour immunofluorescence. Results were compared using
paired t test. RESULTS: Expressed as % +ve in lymphocyte cluster. Mean
+/- 1SD. n = 6. TABULAR DATA, SEE ABSTRACT VOLUME. DISCUSSION: A
decrease in proportion of CD4+ lymphocytes was seen following
cryopreservation. This decrease was not evident in the CD4+CD45RA+ or
CD4+CD28- subsets. Further studies are required to determine whether HIV
infected CD4+ cells have a survival disadvantage during the
cryopreservation process.
